    📖 🔥 New online #firmonertinib in EGFR #PACC-mut NSCLC. @Cancer_Cell 👉 Firmonertinib has a trifluoroethoxy group, which enhances its potency against EGFR PACC mutation 👉 68% response rate with firmonertinib 240 mg daily.     😜 Are we ready for the era of #PD1×VEGF #bispecific immunotherapy? 📖 Now online in #LancetOncology: #HARMONi — #ivonescimab plus chemotherapy in patients with #EGFR-mutant #NSCLC after progression on EGFR TKI therapy. 👉 First global randomized phase 3 study of a PD1×VEGF bispecific antibody N = 438, with 23% of patients enrolled in the United States 👉 Progression-free survival: 6.8 vs 4.4 months, HR 0.52 👉 Overall survival: 16.8 vs 14.0 months, HR 0.79 👉 Encouraging evidence of protection against CNS progression 💡 HARMONi represents an important step in evaluating the potential of simultaneously targeting PD-(L1)1 and VEGF.     📖 Now online @CD_AACR 🔥 👉 The oncogenic #EGFR–SHC1 #fusion drives hashtag#lungcancer through dual activation of the N-terminal EGFR kinase domain and C-terminal SHC1 phosphorylation sites. This biology requires combined inhibition of both EGFR and SRC-family kinases to achieve effective clinical control. 👉 Most importantly, this represents a novel fusion activation mechanism: SHC1 acts not merely as a facilitating #fusion partner, but as a true #collaborating partner in oncogenesis: a previously unrecognized mechanism with direct therapeutic implications. 👉 Could similar cooperative signaling by fusion partners help explain de novo or early resistance in other fusion-driven cancers? 😜 @ElizSMcKenna and Jurgen Wolf (University Hospital Cologne, Germany)

    📖 🔥New online from our group at @UTMDAnderson on ultra-precision oncology in #lungcancer 👉 Analysis from over 22,000 #EGFR-mutant tumor samples identified #PACC mutations in 12% of cases (A) 👉 While classical and #Ex20 mutations more commonly occur as single mutations, PACC mutations are more frequently (66%) observed as compound mutations (B). 👉 PACC/PACC and PACC/classical-like compound mutations function biologically similar to single PACC mutations, NOT classical mutations (C) 👉 Compound PACC mutations demonstrate greater sensitivity to EGFR TKIs than single PACC mutations (D) ⚡ 💡 Largest #EGFR mutation landscape in a Western population. ⚡ 💡 #Compound EGFR PACC Eare biologically analogous to #single PACC. ⚡ 💡 Important insights for drug development of next-generation EGFR TKIs.
