Truly a fun collaboration with @SatoshiTodaLab and @Kosuke_Mizu on the SYMPLE3D project, now out in @emboreports! The synthetic biology approach led to the unexpected finding of gradient input resulting in sharp boundary output, demonstrated in 3D spheroids. A short 🧵 below:

How to make a #morphogen? Find some answers in our paper (fresh from the press!) describing the transformation of GFP into a morphogen that can control growth and patterning of the Drosophila wing. #synbio #devbio

In the #neuraltube, progenitors need to take the right fate at the right time & place to build a functional nervous system. Opposing #morphogen gradients provide #positional information. But how do the cells know the time? Let’s look at gradients in growing tissues. 🧵(2/9)

In summary, #morphogen gradients can not only convey position, but also #time. Gradient-based timers can coordinate #cell #differentiation in space. Coupling with cell-intrinsic timers may yield transient responses also with constant activation - as observed in the #neuraltube.

The same duration along the patterning axis can be obtained via a position-dependent readout threshold for the #morphogen. The further away from the source, the lower the threshold needs to be - similarly to #patterning in space. 🧵(6/9)

We show that non-linear #morphogen decay slightly improves #patterning #precision close to the morphogen source, but the effect is minuscule. Far from the source, non-linear decay leads to imprecise gradients. 🧵(2/8)

🧬Preprint alert! FGF3 isn’t just a paracrine signal, it’s a self-editing morphogen. Fgf3-producing cells ignore parts of their own signal via nuclear repression of select target genes. A built-in asymmetry between senders and receivers! #FGF #Morphogen

Turing patterns in time! Cells must differentiate according to their position within the whole (that's why my brain is just (barely) that). But how do they know *when*? Turns out that Turing patterns, which say 'where', in an expanding system, can code for 'when'. Great work!

How does an #embryo measure #time? It turns out that morphogen gradients can also act as timers. Check out our latest @biorxivpreprint : #development #morphogen #patterning #preprint #cells #tissue 👇Thread 🧵(1/9)

@RCG_Heretic Thanks for the comment, but a small correction: it is not a Turing patterning mechanism, just normal #morphogen gradients with a threshold-based readout (French Flag model).
